Experimental Setup:
1. VERY GENTLY, mix the yeast suspension, but DO NOT SHAKE IT! You can tilt the
tube and/or take up some suspension into a clean pipette and squeeze it back into the
suspension WITHOUT CREATING BUBBLES!
2. Mix 1.5 ml of glucose solution with 1.5 ml yeast suspension in a small plastic beaker.
3. Take up the 3 ml of the mixture into a 5 ml syringe. With the tip of the syringe pointed
upward, draw in 1 ml of air.
4. Allow the yeast-sugar solution in the syringe to incubate at room temperature for 5
minutes.
5. Place a drop of dyed water into a serological pipette by simply dipping the tip into the
dyed water and turning the pipette horizontal. Move the water droplet to the zero mark
(or as close as possible) by tipping the pipette back and forth.
6. Assemble the pipette to the syringe with the rubber tubing. Clamp it to a ring stand using
clothespins or clamps.
7. Note the starting place of your water droplet, then record the volume at least every two
minutes. (Closer time intervals for data collection may be needed as the reaction speeds
up.)
8. Graph the data and calculate the slope of the linear part of the curve
